Drilla Killa is one of the Occupied Sector targets in Borderlands 4, and the marker on the map only tells you half the story. The activity sits on Akela Island, but the arena itself is buried under it, at the end of a cave run that ends in a drop you cannot walk back up.
Quick answer: Fast travel to Akela Island on the far west side of the Bagheera Range, enter the cave system, fight through to the security doors and ammo vending machines, then drop into the flooded cavern below to spawn Drilla Killa.

Open the map and pull the camera to the far west edge of the Bagheera Range. Akela Island is the landmass out there, and the Drilla Killa activity marker sits on it, flagged as an Occupied Sector. Providence Island is the neighboring landmass, so if you are looking at that one, you have gone too far across.

Restock at the Lite Vending Machine before the drop
The last thing you pass before the arena is a Lite Vending Machine, and it is the only ammo source on the route. A full ammo refill runs $425, which is cheap insurance against running dry halfway through the fight.
Sweep the chained crates and containers around this area while you are here as well. Once you jump down, the vending machine is out of reach until the encounter is over.

How to confirm the boss fight has triggered
The map screen flips from the cave layout to the boss arena as you approach the final ledge, which is your signal that you are in the right place. The moment you land in the flooded cavern, Drilla Killa spawns and its name and health bar appear on screen.
If nothing happens, you have not dropped in yet. The encounter does not start from the vending machine level or from any of the upper platforms, only from inside the cavern itself.
Refighting Drilla Killa with Moxxi’s Big Encore
Bosses in Borderlands 4 can be respawned on the spot through Moxxi’s Big Encore machine, which charges a percentage of your cash instead of sending you back through the whole cave run. Some Encore machines show up as soon as the boss dies, while others only appear after you leave the area and return.
In Ultimate Vault Hunter Mode, one of the machines is red rather than white. That version spawns a tougher “Big Encore” variant of the boss, costs Eridium instead of cash, and can only be used once per week, but it carries extra rewards for the trouble.
Once you know the route, the whole approach takes under two minutes, and the vending machine sitting right before the drop makes Akela Island a reasonable spot to sit and farm rather than a one-and-done detour.
