The Mariana’s Awakening update turned Mariana’s Veil into a five-stop errand. Every existing layer of the cave system now has a descendant of Vaelor standing near its rod vendor, and each one wants a specific mutated fish caught with that layer’s own rod before handing over a gauntlet. Collect all five and the questline moves on toward the Obsidian Trench.
Quick answer: Talk to Vaelor at Roslit Bay to open the questline, then complete one mutation quest each for Ignis, Glacius, Tenebris, Luminis and Resonus using the rod belonging to their layer.
Gauntlet checklist by layer
The descendants sit in the same order as the normal descent, so it is far quicker to start at the top of the cave system and work down rather than jumping between layers.

None of these quests can be cleared with a generic rod or a generic catch. Each objective is locked to a mutation and to the rod sold in that layer, so the prep work matters more than the fishing itself.

Mutated catch requirements for the first three gauntlets
Ignis, Glacius and Tenebris all follow the same pattern. Two objectives, one of which usually demands a Perfect Catch, and both tied to fish added to their layer with the update.
Ignis in Volcanic Vents (Volcanic Gauntlets)
At roughly (-3343, -2028, 4076), Ignis faces the Volcanic Rod vendor. His requested catch is two fish designated Perfect Catch Ashen Fortune Charred Coelacanth, plus one Ashen Fortune Obsidian-Plated Piranha; use the Volcanic Rod for all three.

Hand both objectives in at Ignis and he gives you the Volcanic Gauntlets on the spot.
Glacius in Challenger’s Deep (Challenger’s Gauntlet)
Start in Challenger’s Deep and head to (738, -3355, -1529), where Glacius is positioned opposite the Challenger’s Deep Rod. Deliver one Perfect Catch Chilled Glacial Gulper plus three Chilled Hoarfrost Halibut; each fish must be caught with the Challenger’s Rod.

Drinking a Glitched potion before you start makes it far easier to line up the Chilled mutation on four separate catches.
Tenebris in Abyssal Zenith (Abyssal Gauntlet)
Tenebris waits on the third layer, Abyssal Zenith, at around (-1043.4, -1043.7, 84.7), across the Anglerfish Pond. Glide over instead of swimming so the anglerfish do not chew through your health on the way. He wants two Wrath Lumin-Lure Lanternfish and one Perfect Catch Wrath Void-Drifter Jelly with the Rod Of The Zenith.

Return to Tenebris with both objectives ticked off and the Abyssal Gauntlet is yours.
Luminis in the Calm Zone and Resonus in the Veil of the Forsaken
The final two descendants follow the same structure but their exact mutation wording and catch counts should be read straight from the quest dialogue in-game. What is fixed is the fish pool each one draws from, since the update seeded both layers with two new species apiece.
Luminis (Calm Zone)
Luminis is the fourth descendant and sits in the Calm Zone, where the two new catches are Pearl-Plated Pleco and Geode Grouper. There is a boat spawn point in this layer, and using it makes the fishing far less fiddly than casting from the edge.

Resonus (Veil of the Forsaken)
Resonus closes out the chain in the Veil of the Forsaken, the deepest of the original layers. Its two new species are Gargoyle Goby and Wraith-Whisper Ray, and Garbage bait is the one to bring. If you lose track of which objective is still open, the Quest Book tracks the chain under Keepers Guidance.

How to confirm a gauntlet has been awarded
A gauntlet is never granted on the catch itself. You have to walk back to the descendant and speak to them again once both objectives show as complete, at which point the reward is handed over and the layer’s entry in the Keepers Guidance chain stops asking for more fish.
A catch only counts if the mutation prefix matches the objective. An ordinary Charred Coelacanth, or an Ashen Fortune one landed with a different rod, will not register against Ignis’ quest.
What the five gauntlets unlock
The gauntlets are not an end in themselves. They feed into the puzzle stage of Mariana’s Awakening, which starts at the Crevice Wall deeper in the abyss. Solve its riddle and you move on to igniting all five layer gems, one for each of the layers you just worked through.
With every gem lit, the Abyss Gate opens and the Obsidian Trench becomes accessible. That is where the new content actually lives, including the Trench Suppression debuff, the Veiled Charybdis reeling encounter, the Skolopendra boss fight and the Chasm-Stalker Rod. Skipping any one descendant stops the whole chain, so treat the five gauntlets as the gatekeeper they are and clear them in descent order.
