This is a 5-star weapon ascension material from Fontaine. It sits at the top of a four-tier line that runs from Dross, to Sublimation, to Spring, and finally to the Essence. You spend it on the final ascension phases of several Fontaine swords, a catalyst, a polearm, and a bow.
You will find it in your bag under Character Development Items, in the Weapon Ascension Materials category. It cannot be used on characters, only on weapons.
How to get Essence of Pure Sacred Dewdrop
There are two reliable ways to obtain it. You can clear the domain at the highest difficulty for a chance at a direct drop, or you can craft it from lower-tier Dewdrop pieces.
Step 1: Travel to the Echoes of the Deep Tides domain in the Court of Fontaine. It only opens on Tuesday, Friday, and Sunday, so plan your farming around those days.
Step 2: Run the Domain of Forgery: Artisanship at level IV, which has a recommended party level of 80. The Essence has a chance to drop from this tier, while lower levels only give the weaker Dewdrop materials.
Step 3: Claim the rewards at the end for 20 Original Resin. Spending one Condensed Resin doubles the haul from a single clear.

If the drop does not appear after a domain run, check that you cleared Artisanship IV specifically, since the lower tiers do not award the Essence.
If you run out before an ascension, the fastest fix is usually the Crafting Bench rather than waiting for the next open day. Keep a small stock of Springs on hand, and you can mint Essences on demand for whatever Fontaine weapon you are leveling next.
