Grow a Chicken Fighter will not let a chicken produce an Ascension Egg. The item only appears when a Rebirth milestone or an Arena rank pays out, both added with the Arena update.
Quick answer: Collect an Ascension Egg at 10, 100, and 250 Rebirths in the Milestones menu, or receive it at Master V (2,200 Trophies) and Grandmaster I.
Ascension Egg Rewards at Rebirth Milestones
Every Rebirth you finish adds one to a lifetime total. That total is what the Milestones list checks. The game has no Rebirth cap, so the counter keeps rising as long as you keep resetting.
An Ascension Egg is a reward on these published rows.

Other milestone rows can pay coins, eggs, or boosts. Only the counts above are the ones that grant this specific egg.
Ascension Egg Rewards in Arena Ranks
Arena is not a 1v1 duel. You send three chickens into each fight. Wins add Trophies, and Trophies move your Rank. The egg is a rank-up reward, not a random drop from a single match.

You need more than 2,000 Trophies before either of these ranks is in range. Master V is the first copy. Grandmaster I is the larger payout. You know it worked when the rank reward is granted and the egg is in your inventory.
Ascension Egg Restrictions
If the egg is missing, the unlock condition has not fired. These are the cases that never produce it.

Those two tracks are the full set of unlocks. Rebirth totals hand out copies at fixed counts. Arena hands out copies only when Master V or Grandmaster I is reached.
