A mutation roll happens separately from the hatch roll. That means even a Skycrest Rooster can come out mutated, so a common pull is not automatically a wasted egg.

Rebirth is the reliable path because it does not depend on other players. The Arena Update attached reward milestones to your rebirth count, and three of them hand over an Ascension Egg.
Rebirth Stones came in with the same update and stack as you complete rebirths. They feed the Rebirth milestone shop, giving late-game accounts a way to buy Ascension Eggs on a repeatable basis instead of waiting on fixed thresholds.
Route 2: Chicken Arena rank rewards
The Arena is the PvP side of the game. You put together a squad of three chickens, fight other players, and bank trophies. Trophies push you up Arena Ranks, and each rank pays out its own bundle of money, Corn Boost, Money Boost, Hatch Luck, and eggs.

The Arena does not run continuously. It opens every half hour, so trophy climbing happens in windows rather than on demand. If you are chasing Master V, line your play sessions up with those openings instead of logging in at random.
Note: Grandmaster I is a percentile rank, not a fixed trophy number. Holding it depends on where the rest of the ladder sits, which makes the 3-egg payout far less predictable than the Master V threshold.
Which route to pick
If your account is already deep into rebirth cycles, milestones are the cleaner option. The thresholds are fixed, the rewards cannot be contested by other players, and Rebirth Stones keep accumulating in the background while you play normally.
The Arena suits players with a strong three-chicken lineup who want eggs faster than a rebirth grind allows. It is also the only route that can deliver three Ascension Eggs from a single reward, though that requires sitting in the top 1%.
Confirming you received the egg
A granted Ascension Egg lands in your egg inventory and can be opened like any other egg. For the Rebirth route, the milestone entry marks itself as completed once the reward is handed over. For the Arena, the egg arrives when the rank itself unlocks, not when you win an individual match.
If nothing appears after a rebirth, check the milestone count rather than the reward. Milestones fire at exactly 10, 100, and 250, so a rebirth at 99 or 249 pays nothing. In the Arena, trophies below 2,200 leave you short of Master V regardless of win streak.
Those two systems are the full set of sources right now. Because no chicken lays the Ascension Egg and no active code drops one, stockpiling depends entirely on how far you push rebirths and how high you climb the Arena ladder.
